Intronic sequences flank the 3′ and 5′ ends of the coding regions and also separate the leader sequence (LP1) from the main coding region of the V gene. Transgenes were cloned into TCR α or β chain expression vectors that contain TCRα or β constant regions and endogenous promoter elements, using Xma I/Sac II sites for the TCRα and Xho I/Sac II sites for the TCR β transgene.
