FP inserts deeper into the membrane on association with TMD as probed by K SV values measured from NBD quenched by Co 2+. The increased K SV values of NBD labeled to the C-terminus of FP and the decrease in K SV for NBD N-terminally labeled to FP when interacting with TMD can be rationalized by a better alignment of FP on complexing to TMD. The results also support the notion of FP-TMD interaction in the membrane.
