The maltose-binding protein with a stalling signal at the C-terminus served as a substrate for tagging, which was detected by anti-His6 antibodies. Addition of a known standard (tmRNA tagging in the 0 frame) at a 1:5 dilution allows comparison with the natural, 0 frame tagging process.
