PARL-mediated cleavage of PINK1. After passing through the Tim23 complex, MPP cleaves the N-terminal MTS of PINK1 and PARL cleaves PINK1 within the TMD. It is not known how PARL can gain access to the PINK1 TMD during import. Cleaved PINK1 is retro-translocated into the cytosol and constitutively degraded by the proteasome via the N-end rule pathway. PARL-mediated PINK1 cleavage may also be supported by other mitochondrial proteases such as m-AAA and i-AAA
