HS at the temperature range 40–55 °C induces ATP- independent cell death. a SO staining of BY-2 cells heat-shocked for 10 min at 40, 45, 50 or 55 °C and imaged after 6 or 24 h. b Quantification of cell death (% SO-positive cells) in the samples showed that pre-treatment with CCCP provided no protection against cell death and protoplast shrinkage at any of the tested HS temperatures. On the contrary, after prolonged exposure (24 h), CCCP appeared to decrease HS tolerance of plant cells. Staining was performed for no HS and HS treatments according to protocols i and ii, respectively (see “Sytox Orange and FM4-64 staining”). IQR, interquartile range.
