Combinatorial assembly of short ALACATs into long ALACATs. To test the ease of editability and swapping of short ALACATs into long ALACATs, we created a ligation reaction in which there were two choices of short ALACAT at each of four or five positions (a). The reaction products were cloned and a total of 81 clones were selected for DNA sequencing, to establish the composition of the long ALACATs (from either the B or H series) were incorporated (b).
