Experimental setup to characterize promoter and terminator activities. A Test strains (right) contain a CDS-YFP reporter controlled by promoters and terminators of different strengths, plus an mCherry reporter as internal control. Control strains (left) with only the mCherry reporter serve to correct for background when monitoring YFP. B Yeast strains cultivated in rich medium with galactose were assessed for optical cell density (OD), and mCherry and YFP fluorescence at the indicated time points using a plate reader.
