The IgLC sequences P001, P006, P013, P016 and P020 were analysed with respect to the position-dependent probability that they can be cleaved by the cathepsins B and D (brown and blue profiles, see ‘Methods’ section for details). The sequences are then aligned with the peptide fragments actually observed in the proteomics analysis of the purified and dissolved fibrils (black lines below the sequences). The PSSM scores of the sequence regions proximal to where cleavage is observed (red bands) are in all cases significantly larger than the average PSSM scores of the sequences, see Mann–Whitney test on the right-hand side. The horizontal dashed line corresponds to a PSSM score of 1.5, corresponding to a cleavage probability of approximately 81.7%
