Working model. By performing scRNA-seq on crushed and uncrushed DRG neurons from SNC rats, an animal model established as a basis for studying the relationship between evoked neuropathic pain and nerve regeneration after peripheral nerve injury, DRG neurons were matched to 7 known subtypes; among these, the PEP1 neuronal subtype in crushed DRG is particularly interesting. After EDSSP and functional verification, the key gene Adcyap1 was found to have a protective effect on pain and nerve regeneration. This study provides new insights into the intrinsic link between neuropathic pain and axonal regeneration following PNI and offers new molecular targets and ideas for early clinical treatment
