Figure 1

H2O2 stabilizes Lif transcripts in rMC-1 Müller cells. A) rMC-1 cells were serum deprived (SD, circle) for two hours. H2O2 (50 μM) was added immediately (SD + H2O2, square) or at one hour during SD (SD + late H2O2, triangle). B) rMC-1 cells were treated with actinomycin D (ActD, circle) for two hours. H2O2 was added immediately (ActD + H2O2, square) or at one hour during ActD treatment (ActD + late H2O2, triangle). RNA levels of the indicated genes were determined by real-time PCR at 0 hour, 1 hour and 2 hours of treatment and expressed relative to levels at 0 hour. Shown are means ± SEM of N = 4 to 5. One-way ANOVA with Sidak’s posttests were used to compare RNA levels against levels after SD (A) or ActD (B) treatments at one hour and two hours. (*) P <0.05, (**) P <0.01, (***) P <0.001 and (****) P <0.0001. ANOVA, analysis of variance; SEM, standard error of the mean.
