Effects of NLRP3 depletion on different markers of inflammation or oxidative stress in mdx muscles. Tibialis anterior muscles were sampled from WT mice, Nlrp3-KO mice, mdx mice and mdx/Nlrp3-KO mice. Three pro-inflammatory cytokines (IL-1Ī², IL-18 and TNFĪ±), one oxidative stress marker (PRDX3) and one anti-inflammatory cytokine (IL-10) were measured at the mRNA and/or protein levels. mRNA levels were normalized to cyclophilin and presented as the relative expression compared to WT values. Protein quantification was performed by measurement of DAB staining areas on immunochemistry sections (like those shown on Fig. 8), and data were presented as percentages of WT values. Results are means Ā± SEM for eight to ten (mRNA) or six (protein) mice per group. #P < 0.05, ##P < 0.01, ###P < 0.001 vs WT; *P < 0.05, **P < 0.01 for indicated conditions
